Main use: o-phenanthroline and ferrous ions form an orange-red complex under the condition of pH 4~5, and then the iron content is determined by spectrophotometry.

Physical and chemical properties: Monohydrate is a white crystalline powder. Melting point 93-94ºC, anhydrous melting point 117ºC, soluble in 300 parts of water, 70 parts of benzene, soluble in alcohol and acetone.

It can form complexes with a variety of transition metals. Since the complexes formed are chelates, they are relatively stable. The complexes formed with copper and their derivatives can be used as non-oxidative nucleic acid cleavage enzymes because they have certain cutting activity on DNA, and thus have certain anti-cancer activity. Properties: When recrystallized with water, it contains one molecule of crystal water. Melting point 91.5ºC (102ºC). When recrystallized with benzene, it does not contain crystal water, and the melting point is 98~100ºC (117ºC). Boiling point is above 360ºC. Soluble in ethanol, benzene, acetone, insoluble in petroleum ether. It forms complexes with iron, copper, cobalt, nickel and 2,2′-bipyridine, and forms a red complex with Fe2+. It is oxidized with potassium permanganate to obtain 2,2′-bipyridyl-3,3′-dicarboxylic acid. It can be used as a quantitative colorimetric reagent for copper and iron, and as an indicator for titrating iron salts with cerium sulfate; it can also be used as a dye for animal fibers.

In a solution with a pH of 2 to 9, o-phenanthroline reacts with divalent ferrous ions (Fe2+). The reaction is highly selective, and the resulting orange-red complex is very stable, with lgK stable = 21.3 (20°C). Its solution has a maximum absorption peak at 510nm (visible light). Using this color reaction, trace iron can be determined by visible light spectrophotometry.

Although the suitable pH range for the color reaction is very wide (pH = 2 to 9), it is usually measured in a HOAc-NaOAc buffer medium with a pH of 5.
